Maruñak,S.L.; Leiva,L; Garcia Denegri,M.E.; Teibler,P; Acosta De Pérez,O.
A phospholipase A2 has been isolated from Bothrops jararacussu venom from snakes that inhabit the northeast region of Argentina. The present study describes in vivo and in vitro biological activities of phospholipase A2 from B. jararacussu as well as isolation details of its. Venom was obtained by milking of adult snakes which were housing in wood reptile cages of varying dimensions in heated (20-30ºC) rooms. Snakes received a weekly diet of mice and water was available ad libitum for drinking and soaking. The enzyme was purified by gel filtration on a Sephadex G-75 column followed by ion exchange chromatography on a SP-Sephadex C25 column. The major peak belonging to proteins was retained in the cation exchanger and then eluted using a concentration...

Isolation and characterization of a serine proteinase with thrombin-like activity from the venom of the snake Bothrops asper BJMBR
Pérez,A.V; Rucavado,A; Sanz,L; Calvete,J.J; Gutiérrez,J.M.
A serine proteinase with thrombin-like activity was isolated from the venom of the Central American pit viper Bothrops asper. Isolation was performed by a combination of affinity chromatography on aminobenzamidine-Sepharose and ion-exchange chromatography on DEAE-Sepharose. The enzyme accounts for approximately 0.13% of the venom dry weight and has a molecular mass of 32 kDa as determined by SDS-PAGE, and of 27 kDa as determined by MALDI-TOF mass spectrometry. Its partial amino acid sequence shows high identity with snake venom serine proteinases and a complete identity with a cDNA clone previously sequenced from this species. The N-terminal sequence of the enzyme is VIGGDECNINEHRSLVVLFXSSGFL CAGTLVQDEWVLTAANCDSKNFQ. The enzyme induces clotting of plasma...

Maintaining rear-fanged snakes for venom production: an evaluation of mortality and survival rates for Philodryas olfersii and P. patagoniensis in captivity J. Venom. Anim. Toxins incl. Trop. Dis.
Braz,HB; Rocha,MMT; Furtado,MFD.
This study evaluates the mortality and average survival rates of captive female Philodryas olfersii and Philodryas patagoniensis snakes maintained for venom production. Also, two factors likely to reduce captive survival were studied - body condition at admission and seasonality. Mortality peaks occurred during the second month in captivity. More than half the individuals were dead at the end of the third month. This suggests that the first three months in captivity are the most critical in terms of survival and adaptation. Females collected and admitted during spring and summer lived less time than those collected in autumn and winter. As gravidity and egg-laying occur during spring and summer, we suggest that the lower survival rates in these seasons may...

Mechanisms of virus resistance and antiviral activity of snake venoms J. Venom. Anim. Toxins incl. Trop. Dis.
Rivero,JVR; de Castro,FOF; Stival,AS; Magalhães,MR; Carmo Filho,JR; Pfrimer,IAH.
Viruses depend on cell metabolism for their own propagation. The need to foster an intimate relationship with the host has resulted in the development of various strategies designed to help virus escape from the defense mechanisms present in the host. Over millions of years, the unremitting battle between pathogens and their hosts has led to changes in evolution of the immune system. Snake venoms are biological resources that have antiviral activity, hence substances of significant pharmacological value. The biodiversity in Brazil with respect to snakes is one of the richest on the planet; nevertheless, studies on the antiviral activity of venom from Brazilian snakes are scarce. The antiviral properties of snake venom appear as new promising therapeutic...

Molecular cloning of a hyaluronidase fromBothrops pauloensisvenom gland J. Venom. Anim. Toxins incl. Trop. Dis.
Castanheira,Letícia Eulalio; Rodrigues,Renata Santos; Boldrini-França,Johara; Fonseca,Fernando PP; Henrique-Silva,Flávio; Homsi-Brandeburgo,Maria I; Rodrigues,Veridiana M.
Background Hyaluronate is one of the major components of extracellular matrix from vertebrates whose breakdown is catalyzed by the enzyme hyaluronidase. These enzymes are widely described in snake venoms, in which they facilitate the spreading of the main toxins in the victim’s body during the envenoming. Snake venoms also present some variants (hyaluronidases-like substances) that are probably originated by alternative splicing, even though their relevance in envenomation is still under investigation. Hyaluronidases-like proteins have not yet been purified from any snake venom, but the cDNA that encodes these toxins was already identified in snake venom glands by transcriptomic analysis. Herein, we report the cloning and in silicoanalysis of the first...

Protein profile analysis of Malaysian snake venoms by two-dimensional gel electrophoresis J. Venom. Anim. Toxins incl. Trop. Dis.
Vejayan,J; Shin Yee,L; Ponnudurai,G; Ambu,S; Ibrahim,I.
Snake venoms comprise a highly complex mixture of proteins, which requires for their characterization the use of versatile two-dimensional electrophoresis techniques. In the present study, venoms obtained from eight snakes (Ophiophagus hannah, Naja kaouthia, Naja sumatrana, Bungarus fasciatus, Trimeresurus sumatranus, Tropidolaemus wagleri, Enhydrina schistosa and Calloselasma rhodostoma) commonly found in Malaysia were separated based on two independent properties, isoelectric point (pI) and molecular weight (MW). Many differences in snake venoms at the inter-family, inter-subfamily, inter-genus and inter-species levels were revealed. Notably, proteins from individuals of the Viperidae family - Trimeresurus sumatranus, Tropidolaemus wagleri and...

Costa,J. O.; Petric,C. B.; Hamaguchi,A.; Homsi-Brandeburgo,M. I.; Oliveira,C. Z.; Soares,A. M.; Oliveira,F..
Two fibrinogenolytic enzymes, Bothrops alternatus metalloprotease isoform (BaltMP)-I and II, were purified from Bothrops alternatus venom using Diethylaminoethyl (DEAE) Sephacel, Sephadex G-75 and Heparin-Agarose column chromatography. Purified BaltMP-I and II ran as single protein bands on analytical polyacrylamide gel electrophoresis and showed molecular weights of 29000 and 36000, respectively, under reducing conditions in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). BaltMP-II, but not BaltMP-I, displayed blood-clotting activity in bovine plasma, which was about 10-fold higher than that of the crude venom. Both enzymes were proteolytically active against bovine fibrinogen as substrate. When fibrinogen and each enzyme were...

Rattlesnake Crotalus molossus nigrescens venom induces oxidative stress on human erythrocytes J. Venom. Anim. Toxins incl. Trop. Dis.
Meléndez-Martínez,David; Muñoz,Juan Manuel; Barraza-Garza,Guillermo; Cruz-Peréz,Martha Sandra; Gatica-Colima,Ana; Alvarez-Parrilla,Emilio; Plenge-Tellechea,Luis Fernando.
Abstract Background Globally, snake envenomation is a well-known cause of death and morbidity. In many cases of snakebite, myonecrosis, dermonecrosis, hemorrhage and neurotoxicity are present. Some of these symptoms may be provoked by the envenomation itself, but others are secondary effects of the produced oxidative stress that enhances the damage produced by the venom toxins. The only oxidative stress effect known in blood is the change in oxidation number of Fe (from ferrous to ferric) in hemoglobin, generating methemoglobin but not in other macromolecules. Currently, the effects of the overproduction of methemoglobin derived from snake venom are not extensively recorded. Therefore, the present study aims to describe the oxidative stress induced by...

Snake venom derived fibrin adhesive in dog uterine healing: biomechanical study J. Venom. Anim. Toxins incl. Trop. Dis.
Moraes,J. R. E.; Camplesi,A. C.; Correia,P. H. A.; Shimano,A. C.; Ferraro,G. C.; Moraes,F. R..
Healing is a complex process with many interfering factors. The objective of this work was to evaluate regeneration strength of non-pregnant adult dog uterus when snake venom derived fibrin adhesive is used to reinforce hysterorrhaphy. Maximum limit and rigidity were analyzed. Twenty uterine horns from 10 dogs were hysterotomized and distributed into 2 equal groups. Hysterorrhaphy was performed using the Shimieden-Cushing double layer suture. In one group, animals received snake venom derived fibrin adhesive as reinforcement. Although neither variable was significantly different, our results showed higher rigidity values in the adhesive group. This can be attributed to the adhesive’s effect on organ elasticity or to more granulation tissue formed in the...

Snake venom galactoside-binding lectins: a structural and functional overview J. Venom. Anim. Toxins incl. Trop. Dis.
Sartim,Marco A.; Sampaio,Suely V..
AbstractSnake venom galactoside-binding lectins (SVgalLs) comprise a class of toxins capable of recognizing and interacting with terminal galactoside residues of glycans. In the past 35 years, since the first report on the purification of thrombolectin from Bothrops atrox snake venom, several SVgalLs from Viperidae and Elapidae snake families have been described, as has progressive improvement in the investigation of structural/functional aspects of these lectins. Moreover, the advances of techniques applied in protein-carbohydrate recognition have provided important approaches in order to screen for possible biological targets. The present review describes the efforts over the past 35 years to elucidate SVgalLs, highlighting their structure and...

Subproteome of Lachesis muta rhombeata venom and preliminary studies on LmrSP-4, a novel snake venom serine proteinase J. Venom. Anim. Toxins incl. Trop. Dis.
Wiezel,Gisele A; Bordon,Karla CF; Silva,Ronivaldo R; Gomes,Mário SR; Cabral,Hamilton; Rodrigues,Veridiana M; Ueberheide,Beatrix; Arantes,Eliane C.
Abstract Background: Lachesis muta rhombeata is one of the venomous snakes of medical importance in Brazil whose envenoming is characterized by local and systemic effects which may produce even shock and death. Its venom is mainly comprised of serine and metalloproteinases, phospholipases A2 and bradykinin-potentiating peptides. Based on a previously reported fractionation of L. m. rhombeata venom (LmrV), we decided to perform a subproteome analysis of its major fraction and investigated a novel component present in this venom. Methods: LmrV was fractionated through molecular exclusion chromatography and the main fraction (S5) was submitted to fibrinogenolytic activity assay and fractionated by reversed-phase chromatography. The modular nature of bradykinin-potentiating peptides isolated from snake venoms J. Venom. Anim. Toxins incl. Trop. Dis.
Sciani,Juliana Mozer; Pimenta,Daniel Carvalho.
Abstract Bradykinin-potentiating peptides (BPPs) are molecules discovered by Sergio Ferreira – who found them in the venom of Bothrops jararaca in the 1960s – that literally potentiate the action of bradykinin in vivo by, allegedly, inhibiting the angiotensin-converting enzymes. After administration, the global physiological effect of BPP is the decrease of the blood pressure. Due to this interesting effect, one of these peptides was used by David Cushman and Miguel Ondetti to develop a hypotensive drug, the widely known captopril, vastly employed on hypertension treatment. From that time on, many studies on BPPs have been conducted, basically describing new peptides and assaying their pharmacological effects, mostly in comparison to captopryl. After...

The red seaweed Plocamium brasiliense shows anti-snake venom toxic effects J. Venom. Anim. Toxins incl. Trop. Dis.
Silva,Geisiane Alves da; Domingos,Thaisa Francielle Souza; Fonseca,Rainiomar Raimundo; Sanchez,Eladio Flores; Teixeira,Valéria Laneuville; Fuly,André Lopes.
Background Snakebite is considered a neglected tropical disease by the World Health Organization. In Brazil, about 70% of the envenomation cases are caused by Bothrops snakes. Its venom may provoke hemorrhage, pain, necrosis, hemolysis, renal or cardiac failure and even death in victims. Since commercial antivenom does not efficiently neutralize the local toxic effects of venoms, natural products have been tested in order to provide alternative or complementary treatment to serum therapy. Therefore, the present study aimed to evaluate the ability of the seaweed Plocamium brasiliense and its active derivatives to neutralize hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic activities of B. jararaca venom. Methods Specimens of P. brasiliense...
